openCanvas has had Tones for quite a while now, so theoretically you could make (almost) print-ready artwork.
You'll still have to edit/re-save it in Photoshop to have truly 8-bit compliant artwork.
I'm assuming with oC 5.5 (which introduces perspective guides) that it will have regular Guides available for marking out frames.
Currently version 5.1 only has a very basic grid function.

But yea, there's no multi-page editing, so it might be a bit fiddly to produce a whole manga/comic in openCanvas.

If you really want a print-ready application, check out MangaLabo (http://www.portalgraphics.net/en/cl/)/ComiLabo (http://www.portalgraphics.net/cl/)
